Molecular and histopathological study of Citrobacter freundii and Edwardsiella tarda infection in bullfrog (Rana catesbeiana) broodstock

Abstract

Bacterial diseases are responsible for the majority of global economic losses in aquaculture production worldwide, and frogs are susceptible to Gram–negative bacteria that cause septicemia. This study records the biochemical and molecular characteristics and pathological effects of Citrobacter freundii and Edwardsiella tarda infection in breeding female bullfrogs (Rana catesbeiana). For the bacteriological diagnosis, clinically ill breeders with lethargy, anorexia, pale skin, macroscopic hemorrhage lesions on the hind legs, abdominal distension and upper part of the haunches were used. Two bacterial isolates were obtained from the liver, they were characterized as Gram negative, catalase positive and oxidase negative bacilli. An experimental test was carried out Koch’s postulates to confirm. The liver isolates from the bioassay (Koch’s postulates), RcM1 (R. catesbeiana Muestra 1) y RcM2 (R. catesbeiana Muestra 2) were subjected to biochemical identification by a commercial API 20E and molecular using whole genome sequencing. The samples presented a biochemical and molecular profile corresponding to C. freundii and E. tarda. histopathology analysis of the initial organisms and the experiment, they showed severe infiltration of erythrocytes, thrombocytes, formation of nodules with bacterial agglomerates in the center, infiltration of fibroblasts surrounding the nodule, multifocal black spots (melanocytes), multifocal necrosis and loss of normal structure in bullfrogs internal organs, such as the liver, kidney, spleen and intestine.
