Abstract

The objective of this work was to characterize the LepClaI(rs29004487), LepKpn2I (rs29004488), LepHphI (rs29004508) and LepSau3AI (rs29004501) single nucleotide polymorphisms (SNP) of the Leptin gene in Carora sires. Blood and semen samples were collected from 43 bulls born between 2002 and 2013 for DNA extraction; this was obtained from salting out methodologies. Genotyping was carried out through the PCR-RFLP analysis and by an amplification system with four primers, refractory to mutations (TETRA PRIMER PCR). The amplification and digestion products were verified in horizontal electrophoresis in agarose gels. Allelic and genotypic frequencies, observed heterozygosity (Ho) were calculated, and Hardy-Weinberg equilibrium was tested. Allelic and genotypic frequencies were respectively 0.98(A), 0.02(T) and 0.95(AA), 0.05(AT) for rs29004487 polymorphism; 0.59(C), 0, 41(T) and 0.14(CC), 0.53(CT), 0.33(TT) for rs29004488; 0.8(C), 0.2(T) and 0.67(CC), 0.26(CT), 0.07(TT) for rs29004501; 0.94(C), 0.06(T) and 0.88(CC), 0.12 (CT) for rs29004508. No deviations from Hardy-Weinberg equilibrium were observed and the existence of linkage disequilibrium was evident among the majority of SNPs studied. The allelic and genotypic frequencies of the polymorphisms evaluated in the Carora bull population were similar to those obtained in other breeds. This first approach in the characterization of the Leptin gene in Carora cattle will allow to consider the genetic potential of the breed for the production of milk or meat.
